Reports out of St. Louis are grim: three people dead and widespread damage from up to six inches of rain caused by the remnants of Hurricane Ike.
Flooding affected one Route 66 alignment in the St. Louis region, Manchester Road. According to the St. Louis Post-Dispatch:

The bad weather also changed or canceled several events Sunday. The Tour of Missouri cycling race course was modified, and race officials canceled a health and wellness expo.
The story also mentions that the Meramec River, which goes through Valley Park and Route 66 State Park, is also expected to go over its banks.
The Belleville News-Democrat reports here and here of localized flooding in the Illinois communities of Edwardsville, East St. Louis, Collinsville and Hamel.
Spotting flooding is being reported throughout Missouri. I don’t see anything relating to 66, but you probably ought to check a map here before traveling.
